I think Toyota would take the opportunity to showcase a new motor in the Supra. Plus to compete with the Z I would imagine they would go with a larger normally-aspirated engine rather than a turbo. The Supra and Z have always been pretty similar in specification although the Z did switch from I-6's to V6's when the first 300ZX came out. Of course Nissan has totally abandoned the I-6 now, even the new Skylines have V6's. Pretty much any six-cylinder Toyota with sporting aspirations is an inline configuration so my guess is that IF a new Supra comes out then it will remain an inline six.
Thing is, rumor has it the new GS will have a V6. And so will the next IS. Basically that Toyota/Lexus wont make an Inline-6 anymore.

That sucks, cuz my favorite (resonable, not like twin turbo V12) engine is a tubro Inline-6.
