M is full manual. You are in control of the shutter and f-stop. P mode is "Program Mode" which is some preset settings for you to take "ideal" pictures in different settings. I might be wrong about your camera having a Shutter or Aperature Priority mode.

There is no "should" when asking what ISO speed you should use. It all depends on what you want. A low ISO will be less sensative to light. A higher ISO will be more sensative to light. With a low ISO, you get less noise pickup. With a high ISO, you get more noise pickup.
