The best way is to remove the moulding from the door, and paint then, but that's a little too complicated for the average joe (or just don't have the time for it). You're probably gonna end up masking off the rest of the car.

Best to scuff sand it with 400 grit wet/dry sand paper. Clean, use adhesion promoter, primer and paint. A couple of coats of clear should do the trick.
